- Implements the business group strategy, analyzes business results to identify wins and opportunities and creates plans to address gaps all in partnership with district leadership
- Ensures Sales Supervisor understands and can execute business group strategy including those processes and behaviors needed to achieve that strategy, validates that behavioral expectations are met, and is held accountable to the execution of that strategy through their people. Ensures Supervisors have the appropriate knowledge and expertise through Path to Excellence Knowledge Zone requirements necessary for them to provide an excellent customer shopping experience and employee development experience.
- Fulfills Customer Experience Leader (CEL) responsibilities to support business results and customer needs, including validating merchandising readiness and display functionality
- Completes Coach the Coach activities, monthly Leadership Capability Reviews, and quarterly PDGs to support Supervisor development
- Completes Talent and Selection responsibilities in partnership with district leadership to support business group strategies and performs other duties as assigned

- 2 years supervisory/management experience
- 3 years sales experience

Five days before the failed vote to recall Wisconsin Gov. Scott Walker, Rob ?Biko? Baker sat behind the wheel of his Ford Fusion in a working-class neighborhood of Milwaukee?s West Side and watched two of his college-age volunteers knock on doors. They were doing what canvassers for the League of Young Voters, the group Baker serves as executive director, often do before an election: reminding urban, working-class black voters of an upcoming vote and exhorting them to participate.

?Official records show that you are registered to vote. We?re visiting voters like you who care about our community to remind you about the special recall election on June 5!? the canvassers began, after asking to speak with a specific member of the household.? ?It looks like a lot of people will be voting this year in the special recall election, and we hope our community turns out, too!?

Only this time there was something new in the scripts that Baker?s canvassers had affixed to their clipboards. ?Check this out,? they read. ?Have you heard that they?re trying to take away our right to vote?? The canvasser was then supposed to go on and explain what had happened that year in Wisconsin: A law had been passed to require voters to present ID, but a court had intervened to halt its implementation. ?Our power to change laws is directly tied to voting. Imagine if you didn?t have the right to vote,? the script read, before delivering some stage directions: GIVE PERSONAL STORY.

Baker looked out on the scene. ?After 2010, we?ve been on the defensive so much about voter ID,? said the 34-year old Milwaukee native, a former hip-hop promoter who did social media for Snoop Dogg before starting work on a doctorate in history. Founded in 2003 as part of a Bush-era renaissance in liberal organizing, the League of Young Voters had shifted its emphasis to mobilizing specifically urban minorities in six states, relying on the efforts of mostly young, black volunteers. It was a task Baker thought had taken on an unanticipated urgency with the spate of new state laws requiring that citizens present ID cards to vote. ?When I left school I didn?t think my job would be convincing people to get drivers? licenses,? he said.??That?s not why I left graduate school.?

Baker was nonetheless approaching the voter ID question in a rather scholarly way. When it looked as though the Wisconsin law would be enforced in 2012, he wondered if it might present a new opportunity for his group: Could raising hackles over the law?s perceived injustice help to mobilize infrequent voters who might otherwise feel they don?t have a stake in election outcomes? Baker sought out partners to help him design a field experiment that could be administered in the June recall to yield lessons for November. The League of Young Voters ended up partnering with two leaders of Washington?s lefty empirical-electioneering establishment: the New Organizing Institute and the Analyst Institute, which specializes in randomized-control trials to measure the impact of campaign activity. ?I?m like a backyard mechanic who?s got a little bit of schooling,? says Baker, ?but I?m not a political scientist.?

The New Organizing Institute helped create walk lists that included only targets who had been identified by voter data as likely to be African-American, a group the league believed would be the most outraged by new voting laws. The Analyst Institute oversaw the process of randomly assigning Milwaukee?s 318 wards to three separate groups: 104 would get a standard get-out-the-vote canvass, 104 would get scripts with the additional language about voting rights and ID laws, and 104 would be in a control group that was ignored altogether by the league?s canvassers.

The streets of Wisconsin were a muddle that weekend, with canvassers from established institutions (like unions and Democratic Party organizations) mixing with those from pop-up groups founded solely to battle Walker; all were boosted by the arrival of out-of-state volunteers. The experimental design couldn?t control for the activities of these other groups, so the best Baker could do was to train his two dozen paid staffers and their teams of often teenage volunteers to stick closely to their instructions when they headed out in to knock on doors.

Over the summer, it became clear that presenting IDs wouldn?t be a concern to Wisconsin that fall, but that it would be in other states that mattered a lot to Democratic fortunes, like Pennsylvania. As Baker waited for the results of his field experiment to be tabulated, a growing constellation of liberal operatives, strategists, and donors had already reached the conclusion that the new issue would permit them to go on offense, mobilizing the most disaffected corners of their coalition.

The Motor City Business Startup Boom

Andy Didirosi had a hunch in 2012. He felt that his hometown, given up for dead, was about to start a new life. Didorosi, 23 at the time, leased an old industrial building near the city's northern limit. He posted a notice on Craigslist, hoping people would come to his big empty building to share tools and ideas, make stuff, and maybe start a small business or two. The 22,000-square-foot facility he named Paper Street attracted graphic artists, jewelry-makers, Web designers, carpenters, metalworkers, a music publicist, a spice-maker, and a motorcycle mechanic. They paid as little as $99 a month for a work space; Didorosi added to the rental income by refurbishing meat slicers and other equipment from bankrupt supermarkets and selling the appliances to new businesses. The money allowed him to buy three Blue Bird buses and start a jitney service to supplement city bus routes. Now, in 2025, Didorosi runs the thriving Detroit Bus Co., and 20-plus small businesses rent space at Paper Street.

Didorosi and Paper Street are emblematic of the DIY ethic that helped bring Detroit back. "It's about starting a creative revolution instead of an industrial revolution," he says.

A few blocks from Paper Street, a nonprofit called i3Detroit is full of new and refurbished tools and machines?>a CNC mill, a plasma metal cutter, a 3D printer, an oscilloscope, welding torches, a machine shop, a woodworking shop, and a video-editing studio. Detroit native Clover McFadden is a TechTown success story. After graduating from college-prep Renaissance High School on the city's northwest side, she earned a degree from Howard University in Washington, D.C., and dreamed of becoming a doctor. But on a return trip to Detroit she discovered Bizdom, which grooms aspiring entrepreneurs at TechTown. McFadden enrolled, developed a business plan, and successfully pitched investors. Her business, Circa 1837, produces and sells clothing adorned with school logos of the nation's traditionally black universities, such as Howard.

"People told me I was crazy to start a business in Detroit, but I think the exact opposite," McFadden says. "There are so many people here who want to see you succeed."

John Kushigian is one of them. Like McFadden, he was born in Detroit and moved away to go to college and start a career. He had a tech-related job in San Francisco, where he also took up furniture and cabinet making. In 1990, he returned to his hometown. "I'd been wanting to do something to help Detroit, something that involved woodworking," Kushigian says.

One spring afternoon, Kushigian visited Barry Randolph, pastor of the Church of the Messiah. Kushigian's eyes lit up when he saw the spacious church basement, where another Detroiter, Jeff Sturges, had already set up shop for people to work on bicycles, appliances, and computers to screen-print clothing, do Web design, and edit audio and video. Kushigian asked whether he could add carpentry to the mix. "Barry gave me a three-word answer: 'Go for it.'"

Soon people from ages 5 to 88 were flocking to the DIY space, using the equipment Sturges and Kushigian had purchased. Today dozens of people are learning skills to prepare them for the workforce. "A couple of teenagers discovered that our computers had the GarageBand app for simple music production, and they figured it out. Then they started spending 5, 6 hours a day writing lyrics. Now a local Internet radio station is interested in their work," Kushigian says. The young men are on their way.

"Detroit for years, during its decline, had been hoping for a corporate messiah," Kushigian says. "The city finally gave up on that. I'm from Detroit, I escaped Detroit, and yet I came back. Why? Because there's an energy, a lit-up, beefed-up energy, a mental aliveness to Detroit. A lot of people here have a fire burning inside. My feeling is that the messiah is us."

A Buddhist statue brought to Germany from Tibet by a Nazi-backed expedition has been confirmed as having an extraterrestrial origin.

Known as the "iron man," the 24-cm high sculpture may represent the god Vai?rava?a and was likely created from a piece of the?Chinga meteorite?that was strewn across the border region between Russia and Mongolia between 10,000 and 20,000 years ago, according to Elmar Buchner of the University of Stuttgart, and his colleagues.

In a paper published in?Metoritics & Planetary Science, the team reports their analysis of the iron, nickel, cobalt and trace elements of a sample from the statue, as well as its structure. They found that the geochemistry of the artifact is a match for values known from fragments of the Chinga meteorite. The piece turned into the iron man would be the third largest known from that fall.

Given the extreme hardness of the meteorite ? "basically an inappropriate material for producing sculptures" the paper notes ? the artist or artists who created it may have known their material was special, the researchers say. Buchner suggests it could have been produced by the 11th century Ben culture but the exact origin and age of the statue ? as opposed to the meteorite it is made from ? is still unknown. It is thought to have been brought to Germany by a Nazi-backed expedition to Tibet in 1938-39. The swastika symbol on the piece ? a version of which was adopted by the Nazi party ? may have encouraged the 1938 expedition to take it back with them.

"While the first debris was officially discovered in 1913 by gold prospectors, we believe that this individual meteorite fragment was collected many centuries before," said Buchner in a statement. "The Iron Man statue is the only known illustration of a human figure to be carved into a meteorite."

Although this item may be the only known human figure carved into a rock fallen to earth, other meteorites have also been used by many religions across the world. A 15-ton example in North America called the?Willamette meteorite?is sacred to some native Americans, while some have suggested that the Black Stone in the Kaaba in Mecca is a meteorite.

Pentagon calls for steps to prevent sexual assault

Defense Secretary Leon Panetta has ordered all military branches to review policies and procedures in an effort to curb sexual assaults in basic training and to improve how officers and senior enlisted leaders are educated and respond to reported incidents.

The goal of the latest initiatives, Pentagon press secretary George Little said, is more uniform and comprehensive training across all armed forces.


"It is clear that the department must continue to do more to prevent sexual assault, especially in initial military training environments," Little said. "Our newest service members are the most vulnerable and most likely to experience a sexual assault."

Panetta directives, which come less than six months after the Pentagon revised how sexual assaults are reported, includes an assessment of how new recruits are trained, who trains them and oversight of instructors. In addition, the department intends to improve training for prospective commanders on sexual assault prevention and response.

Last year, 3,192 sexual assault were reported across all branches of the military, though the Defense Department says the true number of incidents was closer to 19,000 because most sexual assaults go unreported.

The announcement comes after a number of reports on sexual assaults within the military.

Lackland Air Force Base trainers are under investigation for sexual assaults against 31 recruits. NBC's Jim Miklaszewski speaks with one victim from another case who said she was drugged and raped by a recruiter in Maine, but she felt powerless to fight back.

At Lackland Air Force Base in San Antonio, Texas, at least a dozen instructors were accused of sexually assaulting at least 31 recruits and 35 instructors have been removed from their posts during the investigation. The Lackland cases were considered in the latest initiatives, Little said.

?The Invisible War?,?? a documentary film released this summer, unveiled more chilling stories from service members who describe a pattern of assault, intimidation and retaliation, and a failure by the military to hold perpetrators accountable for their crimes. The film is highly critical of the military, and in particular the Sexual Assault Prevention and Response Office, which oversees Defense Department policy on sexual assaults.

The review of training will also assess the potential benefits of increasing the number of female instructors.

"That review will assess initial training in several areas," Little said, "including the selection, training and oversight of instructors and leaders who directly supervise trainees and officer candidates; the ratio of instructors to students; and the ratio of leaders in the chain of command to instructors." It is to be completed by February 2012.

In April, Panetta ordered that sexual complaints be handled by higher ranked officers -- a colonel or officer of equal rank -- to improve accountability. In the past, a service member?s local unit commander would evaluate charges and decide whether to pursue disciplinary action.?

Service members who report a sexual assault also were given the option to quickly transfer from their unit or installation to get away from an alleged assailant.?

Greg Jacob, policy director for the Service Women's Action Network (SWAN), an advocacy group for women in the military, told NBC News that the latest Pentagon moves are a step in the right direction, but fall short of having unbiased lawyers determine whether sexual assault cases should be prosecuted.

"We're really excited to see the secretary stepping out with leadership, saying 'OK I want to get a snapshot to see what's going in these schools'," Jacob said of the training review. "But as long as commanders are making those decisions you're never going to have completely unbiased dispositions coming out that command -- unless they kick it over to a criminal prosecutor."

World's largest oil trader flouts Iran sanctions

SINGAPORE/BEIJING?- Vitol, the world's largest oil trader, is buying and selling Iranian fuel oil, undermining Western efforts to choke the flow of petrodollars to Tehran and put pressure on Iran's suspected nuclear weapons program.

Vitol last month bought 2 million barrels of fuel oil, used for power generation, from Iran and offered it to Chinese traders, Reuters established in interviews with 10 oil trading, industry and shipping sources in Southeast Asia, China and the Middle East. A spokesman for Vitol declined to comment.

Swiss-based Vitol is not obliged to comply with a ban imposed in July by the European Union on trading oil with Iran because Switzerland decided not to match EU and U.S. sanctions against Tehran.

The company earlier in the year stopped trading Iranian crude oil from its main European offices before the July 1 EU embargo deadline. But the trading sources said it has continued to deal in Iranian fuel oil from the Middle East.

The tale of the cargo of Iranian fuel oil involves tanker tracking systems being switched off, two ship-to-ship transfers, and blending of the oil with fuel from another source to alter the cargo's physical specification.

Privately-held Vitol SA is led by its long-time CEO Ian Taylor, a Briton. Taylor was among leading donors to Britain's ruling Conservative Party named in March by the Prime Minister's office as having dined with David Cameron at his private apartment in Downing Street amid the fall-out from a "cash for access" party funding scandal. Britain is a vociferous critic of Tehran's nuclear program and a leading advocate of the EU sanctions.

Swiss loophole
Vitol has said previously it is in compliance with sanctions against Iran, but has declined to say whether or not it would follow the strict EU regulations rather than Switzerland's.

Rival Swiss-based traders Glencore and Trafigura said in July they had halted all Iranian oil trade, even though the Swiss government opted against following measures imposed by Washington and Brussels.

The measures have halved OPEC member Iran's crude oil export revenues, devaluing the rial currency and bringing financial hardship to millions of Iranians.

On top of the EU ban, Iran's four biggest oil buyers - China, India, Japan and South Korea - have reduced their imports by at least a fifth to secure exemptions from the threat of U.S. financial sanctions on their companies.

Vitol last year earned record revenue of $297 billion, a near-five-fold increase since 2004. It does not reveal profits.

Fuel oil is a small part of its trading portfolio, accounting for $24 billion of revenue last year compared to $105 billion from crude and $100 billion from other refined products.

Profit margins on oil trade are typically very low, but traders said Iran's difficulties in finding buyers because of sanctions are likely to have made for a fat profit margin for trading its fuel oil.

Ship-to-ship
Vitol acquired the Iranian fuel oil early this month in a ship-to-ship transfer off Malaysia from a National Iranian Tanker Company (NITC) vessel, the Leadership, onto a Vitol-chartered tanker, the Ticen Ocean.

The Ticen Ocean was sub-contracted by Vitol for floating storage off the Malaysian port of Tanjung Pelepas from Titan Petrochemicals, a Hong Kong company which itself hired it from shipowner Frontline of Norway.

Frontline said Titan had told it the ship was not used to store Iranian oil. "Our only counterpart in this matter is Titan and they have said it is not correct that there has been Iranian oil on the boat," said Frontline CEO Jens Martin Jensen.

Titan did not return calls or emails seeking comment.

The Leadership left Iran's main oil export terminal at Kharg Island during the week of August 23, passing through the Strait of Malacca before disappearing from freight tracking systems off the Malaysian coast on September 4. Since sanctions were imposed, Iranian vessels have frequently switched off the onboard 'black-box' transponders used in the shipping industry to monitor vessel movements.

Industry sources in Tanjung Pelepas who monitor shipping transfer operations in Malaysian waters said Vitol later brought alongside another tanker, the Speranza, to replace the Ticen Ocean as floating storage. The Speranza, owned by China's Sino Shipping Holdings, arrived at Tanjung Pelepas on September 13-14, Reuters data shows.

Vitol also transferred some of its fuel oil from the Ticen Ocean between September 11-12 to another vessel, the Kamari I, according to Reuters data. That cargo was delivered to Vitol's storage terminal on the Malaysian island of Tanjung Bin, inside Tanjung Pelepas port, one trading source said.

'Special blend'
Traders said the company then blended the oil in storage with fuel oil sourced from Europe, calling it a "special blend" and offered it to Chinese traders. Reuters was given a copy of the specifications of the cargo on offer.

Vitol first asked a $30 premium to Singapore's benchmark 180-centistoke fuel oil price, said a Chinese industry executive who manages some of China's many small, independent refineries, known as teapots. That would have valued the 2-million-barrel cargo at about $250 million.

"Because the offer was too high, our people didn't really carry on the talks," the Chinese executive said. "Vitol also appeared not in a hurry to sell, so was not being aggressive."

Two Asian refinery buyers who were then offered the oil said the asking price was cut to a $12-$14 premium to Singapore benchmark prices, or around $175 million in total.

"Vitol is offering the cargo as a special blend to teapot refiners in Shandong," said a China-based trader. "No one's agreed to buy the Vitol cargo. I declined because I wasn't sure of the quality and specifications."

At the time of publication it is not known whether Vitol had agreed a deal to sell the oil.

Insurance
Vitol's use of the Ticen Ocean to store Iranian oil could put the tanker's insurance at risk. The vessel is insured by the North of England P&I Association.

The EU's oil embargo bans EU insurers who underwrite around 90 percent of the world's tanker fleet from providing cover for ships carrying Iranian oil.

Mike Salthouse, director of North Insurance Management, which acts as manager for the North of England P&I Association, said it would not provide cover for tankers storing or transporting Iranian oil.

"We would not expect to continue to insure any fleet actively engaged in this trade," he said, but declined comment specifically on the Ticen Ocean.

Scolari says he plans to coach again in 2013

SAO PAULO (AP) -Brazilian World Cup-winning coach Luiz Felipe Scolari says he is likely to start managing again in 2013.

He says he expects to "get a coaching job again next year," although without elaborating whether it will be with a club or a national team. The 63-year-old Scolari has said in the past that he wants to manage a national team at the 2014 World Cup in Brazil.

Scolari, who led Brazil to the 2002 World Cup title, and eight-time national champion Palmeiras mutually ended their contract two weeks ago because the coach had failed to get the team out of relegation zone.

Scolari's comments Tuesday came as Brazil's sports ministry announced he will work as a consultant, giving speeches to young Brazilians about his sports experience and promoting the World Cup.

Hamas head Mashaal says he will resign

GAZA CITY, Gaza Strip (AP) ? The supreme leader of the Palestinian Islamic militant group Hamas has decided to step down, clearing the way for the movement to choose a new head for the first time in more than 15 years, two senior officials said Monday.

Khaled Mashaal told a recent meeting in Cairo of Hamas' leadership that he would not run in upcoming elections for the top position, said Izzat Risheq, a confidant of Mashaal who attended the gathering. Moussa Abu Marzouk, Mashaal's deputy, also confirmed the decision.

Mashaal will remain in his post until a new leader is chosen, most likely by the end of the year.

Hamas officials said the leadership change would not affect its confrontational approach to Israel. The group's founding charter calls for the destruction of the Jewish state, and its leaders have staunchly rejected Palestinian peace efforts with Israel.

It may however impact the balance between exiles like Mashaal, who tended to favor reconciliation with other Palestinian groups, and influential members from the group's Gaza Strip stronghold who are more reluctant to give up their grip on power.

Mashaal's announcement comes at a turbulent time for Hamas. Officials said Mashaal was motivated by a number of factors, including the Arab Spring protests jolting the region and the simmering struggle between Gaza and the exiles.

Mashaal has led Hamas since 1996, helping to build the movement into a potent force. Under his leadership, Hamas carried out numerous suicide bombings and other attacks that killed hundreds of Israelis during a Palestinian uprising a decade ago. He also survived an Israeli assassination attack in 1997. The group has been branded a terrorist organization by Israel, the U.S. and the EU.

Forging an alliance with Iran and Syria, where the group has maintained its headquarters, it also has become an important regional political player. The group swept Palestinian legislative elections in 2006.

After a short-lived attempt at sharing power with President Mahmoud Abbas' Fatah movement, Hamas overran the Gaza Strip in 2007. Hamas continues to rule Gaza, while Abbas governs in the West Bank. The two factions have been unable to reconcile.

Risheq said Mashaal told the recent leadership meeting that given the changes sweeping the region, it is time for him to step down and make way for "new blood."

But other Hamas officials say there have been differences between Mashaal and the influential Gaza leadership over key policy issues, particularly in the area of Palestinian reconciliation. Gaza leaders believe Mashaal was too willing to make concessions in attempts to reconcile with Abbas.

Mashaal also may have some personal motives. He is expected to take on a position with Hamas' parent, the pan-Arab Muslim Brotherhood, whose offshoots have grown in power thanks to the toppling of autocrats in the Arab Spring. Egypt's new president, Mohammed Morsi, hails from the group.

He may potentially seek a leadership role in the Palestine Liberation Organization. Hamas has long sought a place in the PLO, which is dominated by Fatah.

The officials spoke on condition of anonymity because they were discussing the internal workings of the group.

Although a successor has not been named, the two leading candidates are Abu Marzouk, who led Hamas in the 1990s until he was imprisoned in the U.S. for two years, and Gaza Prime Minister Ismail Haniyeh.

Both men are from Gaza, a reflection of the rising influence of Hamas' Gaza wing.

(AP) ? A Philippine official said Monday that a legendary collection of shoes and other possessions left behind by former first lady Imelda Marcos and her dictator husband when they were ousted by a 1986 "people power" revolt have no historical significance, except for some Philippine-made gowns.

Communications Undersecretary Manolo Quezon issued the statement after National Museum officials acknowledged that the collection of Marcos' clothes and shoes had been neglected for more than two decades and some had been damaged by a monsoon storm last month.

Museum officials told The Associated Press on Sunday that termites, storms and neglect damaged part of Mrs. Marcos' collection of at least 1,220 pairs of shoes and other items while they were stored from 1986 to 2010 at the Malacanang presidential palace and later at the National Museum in Manila.

More than 150 cartons of clothes, dress accessories and shoes were transferred to the National Museum for safekeeping two years ago from the riverside palace. They deteriorated further at the museum after the boxes were stored in a padlocked room that was inundated by heavy rains last month because of a leak in the ceiling, museum officials said.

Quezon said the contents of the boxes have "no historical significance except some of the clothes" made by prominent Philippine couturiers. Museum officials said the Philippine-made gowns might be displayed in public, but they were wary because of the "politically sensitive nature of their provenance."

A Philippine historian, Michael Xiao Chua, said the Marcos items are important and should have been displayed to remind current and future generations of Marcos' dictatorial rule and encourage them to safeguard the country's democracy.

The damaged items include native see-through barong shirts, which Marcos often wore in public ceremonies during his 20-year rule. AP journalists saw a barong shirt with a presidential seal on top of a box of 100 shirts which had reddish stains and a sleeve that was nearly torn off.

The Marcoses fled the Philippines at the climax of an army-backed "people power" revolt which became a harbinger of change in authoritarian regimes worldwide. Mr. Marcos died in exile in Hawaii in 1989 and his widow and children returned home years later.

They left staggering amounts of personal belongings, clothes and art objects at the palace, including Imelda Marcos' shoes which have come to symbolize her extravagance amid crushing poverty in the Southeast Asian country.

Ferdinand Marcos' successor, democracy icon Corazon Aquino, accused him of stealing billions of dollars during his rule and ordered many of his assets seized.

Aquino had Imelda Marcos' shoes displayed at the presidential palace. They were removed from public view and stored in the palace basement when Aquino stepped down in 1992. About 800 pairs were loaned in 2001 to a shoe museum which has since become a tourist attraction.

Researchers report novel approach for single molecule electronic DNA sequencing

DNA sequencing is the driving force behind key discoveries in medicine and biology. For instance, the complete sequence of an individual's genome provides important markers and guidelines for medical diagnostics and healthcare. Up to now, the major roadblock has been the cost and speed of obtaining highly accurate DNA sequences. While numerous advances have been made in the last 10 years, most current high-throughput sequencing instruments depend on optical techniques for the detection of the four building blocks of DNA: A, C, G and T. To further advance the measurement capability, electronic DNA sequencing of an ensemble of DNA templates has also been developed. Recently, it has been shown that DNA can be threaded through protein nanoscale pores under an applied electric current to produce electronic signals at single molecule level. However, because the four nucleotides are very similar in their chemical structures, they cannot easily be distinguished using this technique. Thus, the research and development of a single-molecule electronic DNA sequencing platform is the most active area of investigation and has the potential to produce a hand-held DNA sequencer capable of deciphering the genome for personalized medicine and basic biomedical research.

A team of researchers at Columbia University, headed by Dr. Jingyue Ju (the Samuel Ruben-Peter G. Viele Professor of Engineering, Professor of Chemical Engineering and Pharmacology, Director of the Center for Genome Technology and Biomolecular Engineering), with colleagues at the National Institute of Standards and Technology (NIST) led by Dr. John Kasianowicz (Fellow of the American Physical Society), have developed a novel approach to potentially sequence DNA in nanopores electronically at single molecule level with single-base resolution. This work, entitled "PEG-Labeled Nucleotides and Nanopore Detection for Single Molecule DNA Sequencing by Synthesis" is now available in the open access online journal, Scientific Reports (2, 684 DOI:10.1038/srep00684, 2012), from the Nature Publication group.

The reported nanopore-based sequencing by synthesis (Nano-SBS) strategy can accurately distinguish four DNA bases by detecting 4 different sized tags released from 5'-phosphate-modified nucleotides at the single molecule level for sequence determination. The basic principle of the Nano-SBS strategy is described as follows. As each nucleotide analog is incorporated into the growing DNA strand during the polymerase reaction, its tag is released by phosphodiester bond formation. The tags will enter a nanopore in the order of their release, producing unique ionic current blockade signatures due to their distinct chemical structures, thereby determining DNA sequence electronically at single molecule level with single base resolution. As proof-of-principle, the research team attached four different length polymer tags to the terminal phosphate of 2'-deoxyguanosine-5'-tetraphosphate (a modified DNA building block) and demonstrated efficient incorporation of the nucleotide analogs during the polymerase reaction, as well as better than baseline discrimination among the four tags at single molecule level based on their nanopore ionic current blockade signatures. This approach coupled with polymerase attached to the nanopores in an array format should yield a single-molecule electronic Nano-SBS platform.

In previous work, the Center of Genome Technology & Biomolecular Engineering at Columbia University, led by Professor Ju and Dr. Nicholas J. Turro (William P. Schweitzer Professor of Chemistry), developed a four-color DNA sequencing by synthesis (SBS) platform using cleavable fluorescent nucleotide reversible terminators (NRT), which is licensed to Intelligent Bio-Systems, Inc., a QIAGEN company. SBS with cleavable fluorescent NRTs is the dominant approach used in the next generation DNA sequencing systems. Dr. Kasianowicz and his group at NIST pioneered the investigation of nanopores for single molecule analysis. They previously reported that different length polymers, polyethylene glycols (PEGs), could be distinguished by their unique effects on current readings in a ?-hemolysin protein nanopores at single molecule level and subsequently developed a theory for the method. Their results provide the proof-of-concept for single molecule mass spectrometry. The combination of the SBS concept with the distinct nanopore-detectable electronic tags to label DNA building blocks led to the development of the single-molecule electronic Nano-SBS approach described the current Scientific Reports article (09/21/2012).

As lead author Dr. Shiv Kumar points out, "The novelty of our approach lies in the design and use of four differently tagged nucleotides, which upon incorporation by DNA polymerase, release four different size tags that are distinguished from each other at the single molecule level when they pass through the nanopore. This approach overcomes any constraints imposed by the small differences among the four nucleotides, a challenge which most nanopore sequencing methods have faced for decades." Moreover, the technique is quite flexible; with PEG tags as prototypes, other chemical tags can be chosen to provide optimal separation in different nanopore systems.

With further development of this Nano-SBS approach, such as the use of large arrays of protein or solid nanopores, this system has the potential to accurately sequence an entire human genome rapidly and at low cost, thereby enabling it to be used in routine medical diagnoses.
